Former Blackpool man Clarke is also delighted with the move, which follows three seasons at Southend.
"It was quite an easy decision to make given the enthusiasm of the manager and chairman," he said.
"The move up here suits my personal circumstances as well, and I can’t wait to get going.
"The manager has seen me play and knows what he will get. I will do a job for the greater good of the team.
"I’m fully committed, I wear my heart on my sleeve, and I’ll give everything I have.
"I’d like to think my positive attitude is coupled with a bit of quality as well."
Southend boss Steve Tilson is disappointed to be losing Clarke, for whom he paid Blackpool £300,000 in 2006, saying: "He’s going to be missed, but Huddersfield are nearer to his home and have made him a fantastic offer which we are unable to match."
