Dutch international John, 24, has been with the Portuguese giants since leaving Twente Enschede in 2012 and was on loan at Reading last season.
He netted in both Championship games against Town (a 2-2 draw at the Madejski Stadium in November and 3-1 Town win in the return in March)
John previously had a stint at German side Hamburg.
Former England Under 19 international Borthwick-Jackson made his Premier League debut as a substitute in the 2-0 home win over West Bromwich Albion last season.
Both could feature for Wolves in Tuesday’s League Cup second-round tie against Cambridge United at Molineux.
Wolves, who are third in the Championship under Italian boss Walter Zenga, two points behind leaders Town, have yet to be beaten this season.
